I will in Tokyo in mid February for 7 days. I am thinking spending 4 day in Tokyo and other day doing some day trip by train to some interesting place within 2 hour train ride. Prefer to have direct connection or maximum 1 transfer as I am travelling with my daughter in light wheelchair. My family is staying at near UENO station.

There are a couple of options to mind. For some ideas have a look at our easy day trips from Tokyo page.
Especially Nikko is a nice visit to make and is only one transfer from Ueno.
You can also head up to Matsumoto. There's a limited express route from Shinjuku, Tokyo there and the ride itself is very scenic to make.

There are many possibilities. Some of the more popular places are Nikko, Kamakura/Enoshima, Kusatsu, Karuizawa, Izu, Hakone, the Fuji 5 Lakes area, Kawagoe and the Boso Peninsula. It is all up to your interests.
