I need some advise I am going to be traveling to Japan in October and would like to know what is the best way to get from Tokyo to Osaka ( I am ending my trip in Osaka).

If you are doing a single leg from Tokyo to Osaka and are not travelling with a JR Pass then you have several options, each a blend of convenience, time and cost:
- Bullet train trip (medium expense, highly convenient, very fast, fun!)
- Discount airline (low-medium expense, low convenience, medium speed, everyone hates airlines!)
- Highway bus (low expense, medium convenience, low speed)
If very much depends on what kind of traveller you are, but I do recommend the travelling on the bullet train at least once if you have not used it before.

Yes - you can purchase bullet train tickets from any large JR station. I recommend visiting Tokyo station on arrival, or even the JR ticket counter at Narita or Haneda airports and making your booking there.
